Remember that this command is in the context of Col 3:16, letting the Word of Christ richly indwell. As we discussed in that comment, being filled with God's Word is closely associated with being filled with God's Spirit. (Colossians 3:22, 23, 24). When we Colossians 3:23-24 instructs us to do our best in all that we do.
